With programmed cell death, DNA fragments are released. The released DNA fragments have preserved the genetic and epigenetic code of the cell of origin. The fragmentation of the released extracellular DNA (eDNA) in the blood plasma reflects the tissue origin of eDNA and the mechanism of its release.

eDNA is an important diagnostic, prognostic or predictive biomarker for various types of cancer: Blood plasma from patients with metastases generally shows higher levels of eDNA than in healthy individuals. eDNA levels decrease in response to successful therapy, while remaining constant or increasing in situations that are associated with tumor recurrence or worsening patient condition. Until now, however, eDNA-based approaches have primarily focused on the analysis of double-stranded DNA (dsDNA).
